How To Use This Catalog
Shipping and ordering information is given in the Service Information section on pages 3

studies indicate that a cold pack is not necessary with most of the reagents we offer,

including our high quality detergents (See page 110). For your convenience, we continue
to ship most of our products by overnight service for our

ucts. Look in this section for general information on detergent and other product areas.
The New Product section on page 24 lists all the products that have been added since
February 2003.
To locate a particular detergent or other reagent, please check the Alphabetical Product
Listing on pages 25-105 or use one of the three indexes available (Catalog Number
Index, pages 117-118; Chemical Name Index, pages 119-123 or Registry Number
Index, pages 124-125). Note that the Chemical Name Index lists a number of
common names for many of our products. A search function is also available at

Product Use
Anatrace products are supplied for laboratory use only. They are to be used for laboratory research and
experiments only. They may not be used for drugs, cosmetics, pesticide products or any use other than

stated above.
Quality
It is our goal to always supply the finest quality products available anywhere. If you are not completely
satisfied that the chemical supplied to you meets the specifications described herein, we will replace it.
Package Weight
Unless otherwise specified, the package will contain at least the indicated amount and usually slightly
more. The user is cautioned to always measure the required amount from the container.

Our research staff will be pleased to work with you on the synthesis and purification of a novel detergent
or other reagent for your individual research needs.
Custom Ampuling and Reagent Preparation
Do you have a reagent that you must prepare frequently? Isn't it a waste of valuable lab time doing such

Should you receive a product ordered from this catalog which is believed to be nonconforming, please
return the material within 30 days. You must receive authorization from Anatrace before returning the
product. Returned products that have been ordered in error or have been retained more than 30 days

are subject to a 25% restocking fee.

Kits
To select the optimum detergent or combination of detergents and reagents for a particular
membrane protein usually requires testing of many detergents. Note also that the detergent
which is suitable for extraction may not be useful for storage of the purified protein or for
studies conducted on the purified protein. To aid in the selection process, Anatrace offers

a wide variety of solid and solution based detergent kits.
Listed below are nonionic and ionic Master Detergent Kits which contain one gram of each
detergent. Following the master kits are several smaller kits which likewise contain one
gram of the specified detergents.

Anatrace detergents are now available in kits as ampuled 10% solutions stored under argon. These

solution kits cost a fraction of the one gram detergent kits, making the selection of the “right” deter-
gent for extraction of a membrane protein less expensive. The master kit listed below contains nearly
every detergent offered by Anatrace that possesses sufficient solubility to prepare a 10% solution.

Nonionic Detergents
Most of the detergents useful in the extraction and purification of membrane proteins are Nonionic detergents which
possess either a sugar based or a polyoxyethylene head group.
Sugar-based detergents
Sugar-based detergents are among the most popular type of detergents used with membrane proteins. The alkyl
glucopyranoside series is available from the six carbon chain tail to dodecyl-β-D-glucopyranoside. The somewhat
less polar thioglycoside analogs of some of these compounds are also available.
The high CMC value of hexyl-β-D-glucopyranoside (0.25M) indicates that a detergent with a shorter chain would
not be useful. Decyl-β-D-glucopyranoside has a reasonable CMC value, but is marginally soluble. Dodecyl-β-D-
glucopyranoside is even less soluble.
For many membrane proteins, a member of the maltopyranoside series is milder and more effective than the compa-
rable member of the glucopyranoside series.

Another advantage of the maltopyranoside head group is the increased solubility of the higher members of this
series compared to the glucopyranoside series. Even hexadecyl-β-D-maltopyranoside is soluble although it must
be heated to 50

o
C for a few minutes to make a “true” solution. The entire series of maltopyranoside detergents
are available from hexyl-β-D-maltopyranoside to hexadecyl-β-D-maltopyranoside, except for the pentadecyl
member.
Since the maltopyranoside detergents are generally milder than the glucopyranoside detergents, one might think
that maltotrioside detergents might be even more compatible with membrane proteins. Detergents with even larger
head groups seem to be impractical because the higher polysaccharides are not available in pure form at a reason-
able price.
Two sugar-based detergents that have been available for some time are sucrose monododecanoate and sucrose
monodecanoate. As with the maltose-based detergent series, these detergents are mild and useful for the extrac-
tion of membrane proteins. The researcher should be aware, however, that these detergents are esters rather than
glycosidic compounds and thus are easily hydrolyzed in acid, base or by proteases. The sucrose ester detergents
are also mixtures of isomers in which the aliphatic ester is linked to any one of the three primary alcohols in the
sucrose structure.

extraction, purification and crystallization of membrane proteins. Support for the

The CMC values for these detergents are higher than for the corresponding linear chains
containing the same number of carbons. Viewed another way, the cyclohexyl tails pack
more hydrophobicity into a shorter effective chain length. Unpublished data from Dr. J. E.
Harlan and Professor R. M. Garavito (Michigan State University) suggest that detergents
that have a shorter chain length, but higher hydrophobicity, are useful for stabilizing proteins
during crystallization.
A study published in late 1999 by the Dana Farber Cancer Institute and Harvard Medical
School showed that from the patented CYMAL
®
group of detergents, CYMAL-5
®
may
be the detergent of choice for separating and purifying G-protein coupled receptors.

Polyoxyethylene Detergents
Most polyoxyethylene detergents used in the early studies of membrane proteins were
developed years ago for large industrial applications. Today an increasing number of well
defined, highly purified detergents, designed specifically for the extraction of membrane
proteins in their active state are available. Detergents such as dodecyl-β-D-
maltopyranoside and octyl-β-D-glucopyranoside have replaced many polyoxyethylene
detergents. Yet for some proteins the polyoxyethylene detergents remain very useful—
particularly for routine extraction procedures.
Polyoxyethylene detergents are available in a variety of structures and under an often
confusing list of trade names, such as TRITON

, GENAPOL

, BRIJ

, etc. In addition to the confusion caused by trade names, industrial
detergents are often a mixture of closely related detergents that may vary from lot to lot.
They may also contain additives and contaminants which result in undesirable effects during
protein extraction. One such contaminant is an oxidizing agent.

These oxidants can
increase in concentration upon aging.
The “aging” of polyoxyethylene detergents results from the tendency of ethers to react with
oxygen to form peroxides. Light accelerates this process.

In samples of polyoxy-
ethylene detergents, hydrogen peroxide and organic peroxides with a variety of structures
may be found.

The concentration of hydrogen peroxide equivalent can be as high as
0.2%.

Conversely, it may be found that the level of peroxides has not increased over time
because as they form they react with more detergent molecules, resulting in the presence of
several undesirable derivatives.

The presence of peroxides during extraction of membrane proteins has resulted in inactiva-

tion, activation and even fragmentation of biological materials.

Sulfhydryl groups are
readily oxidized by peroxides and such oxidation induces aggregation of acetylcholine
receptors.

Adding to the complexity, sulfhydryl oxidation is implicated in the transition
from a conformation which has a high affinity for acetylcholine to a low affinity form. This
transition has been noted during the purification process.

Interference in biochemical analysis can also be caused by peroxides in detergent solu-
tions. Lever has reported that the Kober reagent, used to measure estrogen, reacted with
detergent solutions containing peroxides to give brown solutions even without heating.

Peroxides are probably responsible for the high blanks noted by Heath and Tappel when
measuring lipid peroxides in detergent-solubilized membrane components.

- Polymeric Solubilization Aids
PMAL

-C12 was the first of a series of new polymeric reagents to be offered by Anatrace. PMAL

-C12 and the series that followed
were first prepared and characterized under an NIH-SBIR grant (GMR4460071) and are classified as amphipols. These polymers have
a unique way of solubilizing an integral membrane protein.

Amphipols wrap themselves around the hydrophobic transmembrane
domains of membrane proteins so that the hydrophobic side chains of the polymer stabilize the hydrophobic surface of the protein,
while the polar side chains of the polymer confer aqueous solubility to the protein-amphipol complex.
Recent papers describe the use of PMAL

-C12 for delivering complex integral membrane proteins (IMPs) into a membrane.

Applications can be imagined where it would be desirable to add a purified membrane protein to lipid vesicles or biological mem-
branes under conditions where the protein could spontaneously insert and adopt its functionally active state without lysing the
target bilayer.

These polymers appear to have some of the functions of the naturally occurring molecules called chaperones.

Since they are
synthetic polymers and not naturally occuring, the mechanism of transport may be different than chaperones. We are calling our
synthetic transport aids Sherpas
®
, after the mountain people who transport items in difficult environments.

MTS Reagents
The ability of MTS reagents (Methanethiosulfonate) to react very rapidly and specifically with cysteine groups

allows the quantitative derivatization of transmembrane proteins. The modification of the functional group attached to
the methylthiosulfonate results in a variety of sulfhydryl specific reagents that can probe the structure of a protein. For
example, charged MTS reagents are soluble in water and insoluble in a hydrophobic environment. Reaction of
charged MTS reagents indicates that the cysteine group is probably on or near the surface and not buried in the
interior of the protein or membrane. Other MTS reagents contain a spin labeled or a fluorescent group to probe the
protein/membrane environment.

Using site directed mutagenesis, one can introduce cysteine residues, one at a time, along a continuous stretch of
residues. Modification of residues, particularly with charged MTS reagents, gives information about the accessibility
of the residue in the ion channel. This technique was pioneered by Arthur Karlin and coworkers.

SCAM, or
substituted cysteine accessibility method, uses a small, polar, MTS reagent to modify each expressed mutant. The
accessibility of the particular residue is inferred from the irreversible functional effects of these reagents.

NDSB Reagents
The series of NDSB's has been found to aid in the solubilization of some proteins. All members of the series, which
are zwitterionic over a pH range of 3-12 and are very water soluble

, have the unique ability to reduce protein aggre-
gation while stabilizing protein structure.

Other applications include refolding and renaturation of denatured proteins,
isoelectrofocusing, and capillary electrophoresis. In addition, NDSB compounds are of benefit in some protein
crystallizations.
